Leader is a papain-like cysteine protease that functions as a virulent factor during replication of foot-and-mouth disease virus (FMDV). It is the first protein to be translated and cleaves itself from the nascent viral polyprotein chain. It is known to block different cellular functions by different mechanisms. One of the most characterized cellular targets of Lpro is the cellular protein eukaryotic initiation factor 4G (eIF4G). During infection Lpro cleaves the translation factor eIF4G blocking the synthesis of cellular proteins. This provides a mechanism that favors viral translation.

I am research microbiologist at USDA/ARS in Plum Island Animal Disease Center. My field of study is to understand foot-and-mouth virus (FMDV)-host interactions. Specifically I study the role of the protease Leader during virus replication.
I also have worked in the Department of Molecular Genetics and Microbiology as well as in the Undergraduate Biology Department at Stony Brook University. There, my research focused on the study of HIV, specifically I studied the different mechanisms and pathways that the virus utilizes to successfully exit the cell for further rounds of infection.
Also, I have taught a research course for undergraduates students whose majors are in Biochemistry and Biology. This idea has come from the outreach program called SMART team offered to high school students (MSOE).
The objectives for this course are to provide the students with a portfolio that will increase their chances of getting into a research lab in such a large institution. The course offers basic knowledge of protein biochemistry and structural biology through lectures, journal clubs and usage of computer softwares that allows the visualization of molecules in 3D (RasMol). Students then joined a research lab for couple of weeks where they model a protein of interest as indicated by the principal investigator. The final model of a protein is then printed in a rapid prototyping facility where critical information can be obtained when a physical model is observed. We are hopeful that this course will serve as a filter apparatus for Faculty members when selecting undergraduate students for research projects.
Lpro upon translation frees itself from the growing polypeptide. This self-processing event is believed to occurred intramolecularly due to the presence of a flexible <scene name='User:Gisselle_Medina/Sandbox7_Leader/C_terminal_extension/2'>C-terminal extension</scene> which contributes to self-processing.
CTE is composed of 18 amino acid residues and presumably can fold back into its own active site. <scene name='User:Gisselle_Medina/Sandbox7_Leader/Active_site_mutated/1'>active site</scene>
