Function

Insulin-like growth factor 2 mRNA-binding proteins (IMP1, IMP2, IMP3) are important factors in cell polarization, migration, morphology, metabolism, proliferation and differentiation. IMPs are expressed mainly in embryogenesis[1].

Relevance

IMPs are highly associated with cancer metastasis and the expression of oncogenic factors. IMP1 is protumorigenic factor in liver carcinogenesis[2]. IMP2 is overexpressed and predicts poor prognosis in pancreatic cancer[3]. IMP3 predicts poor prognosis for colorectal adenocarcinoma[4].

Structural highlights

IMPs contain 2 RNA-binding regions, The N-terminal RRM12 and the C-terminal KH domains. The 3D structure of the complex between IMP3 RRM12 domain and the RNA tetranucleotide CCCC shows the π-π stacking of RNA with Phe and Tyr side-chains of IMP3[5].
