AD1 is a familial early-onset form of Alzheimer disease. It can be associated with cerebral amyloid angiopathy. Alzheimer disease is a neurodegenerative disorder characterized by progressive dementia, loss of cognitive abilities, and deposition of fibrillar amyloid proteins as intraneuronal neurofibrillary tangles, extracellular amyloid plaques and vascular amyloid deposits. The major constituent of these plaques is the neurotoxic amyloid-beta-APP 40-42 peptide (s), derived proteolytically from the transmembrane precursor protein APP by sequential secretase processing. The cytotoxic C-terminal fragments (CTFs) and the caspase-cleaved products such as C31 derived from APP, are also implicated in neuronal death. Defects in APP are the cause of cerebral amyloid angiopathy APP-related (CAA-APP) [MIM:605714. A hereditary localized amyloidosis due to amyloid-beta A4 peptide(s) deposition in the cerebral vessels. The principal clinical characteristics are recurrent cerebral and cerebellar hemorrhages, recurrent strokes, cerebral ischemia, cerebral infarction, and progressive mental deterioration. Patients develop cerebral hemorrhage because of the severe cerebral amyloid angiopathy. Parenchymal amyloid deposits are rare and largely in the form of pre-amyloid lesions or diffuse plaque-like structures. Publication Abstract from PubMed Characterization of amyloid beta (Abeta) oligomers, the transition species present prior to the formation of Abeta fibrils and that have cytotoxicity, has become one of the major topics in the investigations of Alzheimer's disease (AD) pathogenesis. However, studying pathophysiological properties of Abeta oligomers is challenging due to the instability of these protein complexes in vitro. Here, we report that conformation-restricted Abeta42 with an intramolecular disulfide bond at positions 17 and 28 (SS-Abeta42) formed stable Abeta oligomers in vitro. Thioflavin T binding assays, nondenaturing gel electrophoresis, and morphological analyses revealed that SS-Abeta42 maintained oligomeric structure, whereas wild-type Abeta42 and the highly aggregative Abeta42 mutant with E22P substitution (E22P-Abeta42) formed Abeta fibrils. In agreement with these observations, SS-Abeta42 was more cytotoxic compared to the wild-type and E22P-Abeta42 in cell cultures. Furthermore, we developed a monoclonal antibody, designated TxCo-1, using the toxic conformation of SS-Abeta42 as immunogen. X-ray crystallography of the TxCo-1/SS-Abeta42 complex, enzyme immunoassay, and immunohistochemical studies confirmed the recognition site and specificity of TxCo-1 to SS-Abeta42. Immunohistochemistry with TxCo-1 antibody identified structures resembling senile plaques and vascular Abeta in brain samples of AD subjects. However, TxCo-1 immunoreactivity did not colocalize extensively with Abeta plaques identified with conventional Abeta antibodies. Together, these findings indicate that Abeta with a turn at positions 22 and 23, which is prone to form Abeta oligomers, could show strong cytotoxicity and accumulated in brains of AD subjects. The SS-Abeta42 and TxCo-1 antibody should facilitate understanding of the pathological role of Abeta with toxic conformation in AD.
